After mice showed stable orienting, the effects of attention-modulating drugs, MPH and ATX, were assessed. MPH, ATX and saline were administered in a pseudo-randomised cross-over design with a 3- to 4-day washout period between each administration. Mice were subjected to a minimum of two consecutive days of baseline training to ensure continued stable performance between each probe. Mice not reaching criteria (>70% accuracy or completion of 120 trials, excluding anticipations errors) were subjected to further baseline sessions until criteria were met. With a range of 100Hz to 90,000Hz compared to a human’s of 20Hz to 20,000 Hz, mice have excellent hearing and will hear you coming long before you see them. The OB is a bulb-shaped nervous tissue, in humans being located on the ventral anterior aspect of the forebrain in a region known as the olfactory surface, while it is located anterior to the forebrain in mice. This small brain structure is responsible for relaying all olfactory information received by OSNs to the OC. Unlike other sensory systems, olfactory information bypasses the thalamus before reaching its primary sensory cortex; the OB functions as the relay center for olfactory information and directly transmits the information to the various components of the OC including the anterior olfactory nucleus, olfactory tubercle, piriform cortex, and entorhinal cortex [ 28, 29]. Although in humans the OB makes up only ~ 0.01% of the brain by volume [ 30], rodents rely more heavily on olfaction as the rodent OB makes up ~ 2% of the brain by volume [ 31].

Mice must be acclimated to handling for a period of two-four weeks before the onset of intranasal dosing. Acclimation to handling is important, as it helps ensure a correct body position for maximum effectiveness of awake intranasal drug delivery. In addition, mice that have not been properly accustomed to this process can have a severe anxiety reaction after dosing. Mice should spend about 2-3 days on each of nine steps before progressing to the next step, depending upon the animal's comfort to handling. The mouse's stress level should be used as a measure of progress. This means monitoring the mouse's movements, the amount/frequency of urination, defecation, trembling, and biting. If a mouse has a high stress response, stay on that step before progressing to the next until the response is reduced. A sample acclimation schedule can be seen in Table 1. Acclimation of the mice should progress through the following once-a-day steps. Performing these steps more than once per day, in an attempt to accelerate the acclimation, is not recommended. Becoming wobbly when walking about. Always keep an eye on your mouse if they’re housed in a mesh cage. Elderly mice can get caught in the bars if their nails are overgrown. Mice do not have the best vision. It is blurry, though they do have good light and dark perception, with limited color seeing ability.
